牛膝多糖对仔猪淋巴细胞增殖作用和细胞因子分泌量的影响 被引量:26

Effects of Achyranthes Bidentate Polysaccharide on the Proliferation of Lymphocytes and Concentrations of Cytokine in Peripheral Blood Mononuclear Cells of Piglets

摘要 本文旨在研究牛膝多糖(ABP)对仔猪淋巴细胞增殖作用和细胞因子分泌量的影响,探讨ABP免疫调节作用机制。选用128头28日龄的杜长大(杜洛克X长白X大白,Duroc×Landrace×Yorkshire)断奶仔猪,随机分为4个处理,每个处理4个重复,每个重复8头猪。对照组饲喂基础日粮,Ⅰ~Ⅲ组为试验组,分别饲喂在基础日粮中添加ABP500、1000、1500mg/kg的试验日粮。试验第14和28天每个处理随机选取4头仔猪进行前腔静脉采血,测定仔猪外周血淋巴细胞转化率和血清中细胞因子TNF、IL-1β、IL-2、IL-6的含量。结果表明:与对照组相比,试验第14和28天时Ⅰ、Ⅱ、Ⅲ组的淋巴细胞sI值分别提高了8.04%(P〉0.05)和11.32%(P〈0.01)、24.11%(P〈0.01)和19.81%(P〈0.01)、69.64%(P〈O.01)和43.40%(P〈0.01),各试验组之间的SI值差异显著或极显著(P〈0.05或P〈0.01)。与试验第14天相比,各试验组第28天的SI值均呈现下降趋势。试验第14天时,Ⅱ、Ⅲ组细胞因子TNF、IL-1β、IL-2和IL-6含量极显著高于对照组(P〈0.01),Ⅰ组的TNF和IL-2含量显著高于对照组(P〈0.05),但IL.1p和IL.6含量与对照组相比没有显著性差异(P〉0.05);试验第28天时,Ⅰ、Ⅱ、Ⅲ组TNF、IL-1β、IL-6含量显著或极显著高于对照组(P〈0.05或P〈0.01),UI组IL-2含量显著高于对照组(P〈0.05),但Ⅰ、Ⅱ组IL-2含量与对照组没有显著性差异(P〉O.05)。与试验第14天相比,各试验组第28天的TNF和IL-1β含量有增加的趋势,IL-2和IL-6含量有降低的趋势。各试验组之间以Ⅲ组的淋巴细胞转化率和细胞因子的含量增加效果显著。以上结果表明:ABP可促进仔猪血清淋巴细胞转化率和细胞因子TNF、IL-1β、IL-2、IL-6的分泌量。 The experiment was conducted to study the effects of diets added with achyranthesachyranthes bidentatebidentate polysaccharide (ABP) on the proliferation of lymphocytes and concentrations of cytokine in peripheral blood mononuclear cells of piglets and explore the immunity modulation mechanism of ABP. One hundred and twenty-eight 28-day-old three-line crossbred (Duroc × Landrace × Yorkshire) weaning piglets were randomly divided into 4 treatments and each treatment had 4 replicates with 8 piglets per replicate. The control group was fed with basal diet and the test groups (group Ⅰ - Ⅲ ) were fed with basal diet added With 500, 1 000, 1 500 mg/kg of ABP respectively. On the 14th day and 28^th day, 4 piglets of each treatment were randomly picked out for blood-collection from thorax cavity vein, so as to determine lymphocyte proliferation of peripheral blood and the concentration of cytokine TNF, IL-1β, IL-2, IL-6. The results showed that: compared with control group, stimulation indices (SI) in group Ⅰ , Ⅱ , Ⅲ were increased by 8.04% (P〉0.05) and 11.32% (P〈0.01), 24.11% (P〈0.01) and 19.81% (P〈0.01), 69.64% (P〈0.01) and 43.40% (P〈0.01) on the 14^th day and 28^th day respectively and there were significant differences in experimental groups (P〈0.05 or P〈0.01). On the 28th day, the value of SI was decreased compare with that on the 14'h day. On the 14'h day, compared with control group, the concentrations of cytokine TNF, IL-1β, IL-2, IL-6 in group Ⅱ and Ⅲ were significantly increased (P〈0.01). The concentration of TNF and IL-2 in group Ⅰ was higher than that in control group (P〈0.05), and there were no significant difference for IL-1β and IL-6 between the control group and group Ⅰ (P〉0.05). On the 28^th day, the concentrations of TNF, IL-1β and IL-6 in group Ⅰ , Ⅱ, Ⅲ were significantly higher than that in control group (P〈0.05 or P〈0.01); The concentration of IL-2 in group Ill was significantly higher than that in control group (P〈0.05). However, there were no significant difference for IL-2 among group Ⅰ , Ⅱ and control group(P〉0.05). On the 28th day, the concentration of TNF and IL-1β was increased, and the concentration of IL-2 and IL-6 was decreased compared with that on the 14th day. Among the experimental groups, the proliferation of lymphocytes and concentrations of cytokine of group Ⅲ was significantly improved. In conclusion, ABP can improve lymphocyte proliferation and increase the concentration of cytokine TNF, IL-1β, IL-2, IL-6 in peripheral blood mononuclear cells of piglets.
