摘要
对7种大孔吸附树脂的铁筷子总皂苷吸附性能进行了比较筛选试验,并对选定的树脂进行了吸附与解吸优化条件研究。其中LSA-30型大孔吸附树脂具有较佳的吸附能力,其吸附分离铁筷子总皂苷的适宜条件为:上样浓度60 mg/mL,吸附流速2.0 BV/h,洗脱剂为体积分数30%-95%的乙醇,洗脱速度1.0 BV/h,[NaCl]为0.5 mol/mL;95%乙醇洗脱液干燥后总固形物中铁筷子总皂苷含量达到60%以上。
Seven macroporous adsorption resins were compared on their adsorption properties for total saponins from Helleborus thibetanus, and the LSA-30 type resin possessed the better adsorption ability. The optimized conditions of adsorption and desorption were as following: the concentration of sample was 60 mg/mL;the current velocity of adsorption was 2.0BV/h ; the eluting reagent was 30%-95% EtOH ; eluting rate was 1.0 BV/h ; [NaCl] was 0.5 mol/mL. The content of total saponins was up to 60% in the dried fraction of 95% ethanolic elution.
