摘要
目的:观察中药加味补肝汤对糖尿病大鼠坐骨神经VEGF表达的干预作用,探讨其对实验性糖尿病大鼠周围神经病变的防治作用。方法:采用链脲佐菌素诱发糖尿病模型,治疗组予加味补肝汤,分别于治疗后第4、8周末2个时间点用MS302多媒体生物信号记录分析系统测定神经传导速度,免疫组织化学法检测坐骨神经中血管内皮细胞生长因子(VEGF)蛋白的表达,通过MoticImagesAdvanced彩色图文分析系统分析结果。结果:模型组和加味补肝汤组大鼠的神经传导均明显慢于正常对照组(P<0.05),加味补肝汤组快于模型对照组(P<0.05)。在4、8周末模型对照组坐骨神经中VEGF阳性的表达比正常对照组增多(P<0.05),经加味补肝汤治疗4周后VEGF的阳性表达比模型组表达明显增多(P<0.05),治疗8周后比模型组减少(P<0.05)。结论:加味补肝汤对糖尿病大鼠周围神经病变有一定的防治作用,其机制与调节VEGF表达有关。
Objective:To observe the intervene effects of jiawei bugan decoction on VEGF expression in sciatic nerve of diabetic neuropathy rats,explore the preventive and therapeutic effects of jiawei bugan decoction on peripheral neurophathy of diabetes.Methods:Models of diabetes were made by ip 1% streptozotocin,treatment group was given bugan decoction.The nerve conduction speed was recorded by media biosignal system,the expression of VEGF protein in sciatic nerve was tested by immunohistochemistry assay at 4th and 8th weekends.Results:The nerve conduction speed of rats in model and bugan decoction groups was obviously slower than that of normal,the nerve conduction speed of rats in bugan decoction group was faster than that of model.The expression of VEGF protein in sciatic nerve of model rats at 4th and 8th weekends was more than that of normal,which was increased in bugan decoction group at 4th weeken,but dereased at 8th weekend.Conclusion:Jiawei bugan decoction can prevent and cure the peripheral neurophathy of diabetes,regulating the VEGF expression maybe one of its mechanism.
