目的:探讨右旋柠檬烯(d-limonene)诱导人白血病细胞凋亡的作用机制。方法:以人白血病细胞株K562和HL-60为研究对象,采用MTT法及锥虫蓝染色法检测d-limonene对细胞增殖的影响,AnnexinⅤ/PI双重染色法检测其对细胞凋亡的影响,Western印迹法检测线粒体凋亡途径相关蛋白的表达。结果:d-Limonene作用后K562、HL-60细胞凋亡率呈时间及剂量相关性增加;且处理组Bcl-2/Bax比值下降,细胞质中细胞色素c表达增高,caspase-9表达以及caspase-3分裂明显增高。结论:d-limonene能够诱导人白血病细胞凋亡,线粒体凋亡途径的激活可能是其凋亡诱导的主要机制。
Objective:To investigate the mechanism for the apoptosis-inducing effect of d-limonene on human leukemia cells. Methods :This study investigated the effects of d-limonene on the proliferation of K562 and HL-60 cells using MTT assay and trypan blue staining method. Apoptosis was evaluated by annexin V/PI double-staining. Flow cytometry and Western blotting were performed to evaluate expression of the mitochondria-apoptosis pathway-related proteins. Results : d-limonene induced apoptosis of K562 and HL-60 ceils in a dose- and time-dependent manner. The inhibitory effects reached the peak at 72 h (71.21%) after treatment with d-limonene at 0.4 mmol/L. The ratio of Bcl-2/Bax decreased from 1.93 to 0.75. The release of cytochrome c from mitochondria increased of caspase-9 and cleavage of caspase-3 were elevated. Conclusion:d-Limonene induces apoptosis of human leukemia cells. The activation of mitochondria apoptosis pathway may be the main mechanism for the effects of d-limonene on apoptosis of human leukemia cells.
