采用析因设计分析时间和温度对血中谷胱甘肽过氧化物酶活力的影响 被引量:1

摘要 目的采用析因设计分析时间和温度对血中谷胱甘肽过氧化物酶(glutathione peroxidases,GPx)活力的影响,为调查人群GPx活力水平提供血样保存的最佳条件。方法按照析因设计将保存时间(3个水平)和温度(3个水平)进行全面组合,共获得9种组合条件,将大鼠尾静脉血按照9种组合条件进行保存,把当天检测结果作为对照组。采用二硫双硝基苯甲酸(DTNB)法测定GPx活力。结果经析因方差分析,温度对GPx活力无影响(F=2.320,P>0.05),时间对GPx活力的影响有统计学意义(F=15.559,P<0.01),时间和温度之间无交互作用(F=0.921,P>0.05);当血样保存在液氮和4℃时,3、7、14d的GPx活力无变化。而在-80℃保存3天时(47.456±15.444)U/L与对照组(63.048±23.071)U/L比较有差异(t=2.101,P<0.05)。结论血样保存在液氮和4℃时对GPx活力无影响,保存在-80℃时对GPx活力影响显著。 Objectives Using factorial design to analyze the effects of time and temperature on blood glutathione peroxidase( glutathione peroxidases, GPx) activity, in order to provide the best preservative condition of blood samples for surveying GPx activity level in people. Methods According to factorial design, time( three levels) and temperature ( three levels) have been combined into nine kinds of combination conditions, and kept the blood of the rat tail vein in them. We take the immediate test results as the control group. Using dithio - bis - nitrobenzoic acid(DTNB) method to assay GPx activity. Results With analysis of variance of factorial design, the temperature on the GPx activity had no effect ( F =2. 320, P 〉0.05), the time on the GPx activity had a statistically significant effect( F = 15. 559, P 〈 0.01), between time and temperature without interaction ( F = 0.921, P 〉0.05). When the blood samples stored in liquid nitrogen and 4 ℃, the GPx activity did not change at 3 , 7 and 14 days. Compared with the control group (63. 048±23. 071) U / L, while the blood samples stored in -80 ℃and 3 days, GPx activity was lower (47. 456 ± 15. 444) U / L ( t = 2. 101, P 〈 0.05). Conclusions The GPx activity of blood samples stored in liquid nitrogen and 4 ℃ was not changed, when stored in - 80℃ the GPx activity had been affected significantly.
