胃癌组织中livin与caspase-9、p53的表达及相互关系

摘要 目的研究凋亡蛋白livin和caspase-9、p53在胃癌组织中的表达及与胃癌生物学行为关系。方法采用免疫组化S-P检测58例胃癌组织及正常胃黏膜中livin和caspase-9、p53的蛋白表达。结果胃癌组织中livin和caspase-9的、p53蛋白表达阳性率分别为65.5%、20.7%、58.6%,与正常胃黏膜蛋白表达阳性率(分别为3.5%、62.1%、5.2%)比较差异有统计学意义(P<0.05);livin在组织分化差、淋巴结转移者、临床分期高表达率均增高差异有显著性(P<0.05);livin表达与caspase-9蛋白表达呈负相关性(rs=-0.195,P<0.05)。livin与p53蛋白表达呈正相关性(rs=0.851,P<0.05)。结论livin和caspase-9参与胃癌的发生、发展,可能成为胃癌治疗的新分子靶点。抑癌基因p53的失活与凋亡抑制基因livin在胃癌的发生方面可能有相互促进作用。 Objective To study the expression of livin and its relationship with expression of caspase-9 and p53 in gastric carcinoma tissues. Methods Immunohistochemical S-Pmethod was used to detect the expression of livin and caspase-9, p53 proteins in gastric carci- noma tissues and normal gastric mucosas tissues. Results In gastric carcinoma tissues, the positive expressive rate of livin (65.5% vs 3.5% ), caspase-9 (20. 7% vs 62. 1% ) and p53 (58. 6% vs 5.2% ) was respectively significantly higher or lower than that in normal gastric mucosas ( all P 〈 0. 05 ). The expression of livin was inversely correlated with the histologic differentiation of the tumor, lymph node metastasis and clinical stage ( P 〈 0.05 ). A significant negative correlations were observed between the expression of livin and easpase-9 protein in gastric carcinoma tissues ( rs = - 0. 195, P 〈 0. 05 ), livin protein expression was positively related to p53 protein expression in gastric car- cinoma tissues (rs = 0. 851 ,P 〈 0. 05 ). Conclusions Aberrant expression of livin and caspase-9 may be a new target for diagnosis, porgnosis assessment and gene treatment of gastric carcinoma. The aberrant expression of livin and inhibiting carcinoma gene p53 may play facilitative roles in process of carcinogenesis of gastric carcinoma.
