摘要
应用膜片钳全细胞记录技术观察当归提取液(EAS)对酶解分离的单个豚鼠心室肌细胞膜钠通道电流(INa)、L型钙通道电流(ICa-L)的影响。当EAS为132,116和18时,使INa峰值(INamax)从6.98±1.32nA分别降至5.77±1.33,5.28±1.15和3.82±1.45nA(n=6,P<0.01);使ICa-L的峰值(ICa-Lmax)从1005.0±196.5pA分别降至880.1±105.8,597.0±110.5和364.2±98.3pA(n=6,P<0.01)。EAS使INa、ICa-L的电流-电压曲线上移,但均不改变其激活、峰值和反转电位。以上结果表明EAS对INa和ICa-L具浓度依赖性阻滞作用,这可能是EAS具有某些药理效应的离子基础。
Whole cell patch clamp technique was used to investigate the effect of extractant of Angelica Sinensis Diels (EAS) on sodium current (I Na ) and L type callium current (I Ca L ) in enzyme isolated guinea pig ventricular myocytes.EAS at 1∶32,1∶16,1∶8 decreased peak I Na (I Na max ) from 6 98±1 32 nA to 5 77±1 33 nA,5 28±1 15 nA and 3 82±1 45 nA( n =6, P <0.01);decreased peak I Ca L (I Ca L max ) from 1 005 0±196 5 pA to 880 1±105 8 pA,597 0±110 5 pA and 364 2±98 3 pA( n =6, P <0.01).EAS shift up both the I V curve of I Na and that of I Ca L while their active,peak and reverse potentials didnt change.It reveals that EAS blocks both I Na and I Ca L in a concentration dependent manner,which may contribute to some of its pharmacological effect.
