P504s/P63/34βE12一次性标记在前列腺癌穿刺标本中的诊断价值

摘要 目的通过一次性标记的方法,探讨混合型抗体P504s/P63/34βE12在前列腺癌穿刺标本中的诊断价值。方法选择前列腺穿刺标本50例,包括前列腺癌40例,良性前列腺增生10例。同时采用双酶标记法检测混合型抗体P504s/P63/34βE12及单酶标记法分别检测单标抗体P504s、P63及34βE12,观察两种方法的标记结果。结果在40例前列腺癌中,用P504s/P63/34βE12双酶标记法有37例(92.50%)P504s阳性表达,P63、34βE12全部阴性表达;用单酶标记法有36例(90%)P504s阳性表达,P63、34βE12全部阴性表达;10例良性前列腺增生中,两种方法结果完全一致,P504s全部病例阴性表达,P63、34βE12全部病例阳性表达。两种方法在前列腺穿刺标本中表达的一致率为98%。结论通过对两种染色方法的比较,混合型抗体P504s/P63/34βE12双酶标记法较单标抗体单酶标记法更易于观察分辨,尤其对前列腺穿刺标本,前列腺癌的检出率更高,但差异无显著性。 Objective To assess the value of P504s/P63/34βE12 cocktail in diagnosis of prostate carcinoma by one-time staining. Methods Prostate puncture biopsy specimens from 50 patients were selected, including 40 cases of prostate cancer and 10 cases of benign prostatic hypertrophy (BPH). Each of three monoclonal antibodies and cocktail antibodies were used to measure p504s, p63, 3βE12 in different prostate lesions, respectively. The staining results were recorded. Results Of 40 prostatic cancer eases, 37 cases (92.50 % ) stained positively with P504s and all 40 cases stained negatively with P63 and 34βE12 by cocktail- staining; 36 cases (90%) stained positively with P504s and all 40 cases stained negatively with P63 and 34βE12 by monoclonal antibody - staining. Of 10 BPH cases, the same results were obtained by the two staining techniques, all cases stained negatively with P504s, and all cases stained positively with P63 and 34βE12. The agreement rate was 98 % by the two techniques in prostate puncture biopsy specimens. Conclusion Immuno - histochemical analysis with a triple - antibody cocktail (P504s/P63/34βE12) has higher sensitivity and specificity than single antibody staining, and it can improve the detection rate of small focal prostate carcinoma, but there was no statistical difference.
