摘要
目的观察和探讨灵芝孢子粉对戊四氮(PTZ)致痫大鼠脑核转录因子-κB(NF-κB)免疫反应性的影响并进行行为学观测。方法用亚惊厥剂量的PTZ复制大鼠慢性癫痫模型,用药组以灵芝孢子粉灌胃,记录癫痫发作的潜伏期及持续时间,采用免疫组织化学法检测脑NF-κB/P65的蛋白表达。结果大鼠海马和皮质区每高倍视野(×400)下NF-κB/65蛋白表达模型组明显高于对照组(P<0.01);在造模第17、21、25天时用药组较模型组癫痫发作的潜伏期有所延长(P<0.05或P<0.01),海马和皮质区NF-κB/P65蛋白表达用药组低于模型组(P<0.01或P<0.05)。结论NF-κB可能参与了PTZ致痫的发生发展过程,灵芝孢子粉能明显抑制NF-κB蛋白表达,抑制免疫炎症反应,降低神经系统兴奋性,对神经细胞具有保护作用。
Objective To investigate the effects of ganoderm lucidum spores powder on the behaviors and immune reactivity of brain nuclear factor-κB(NF-κB) in rats with epilepsy induced by pentetrazole (PTZ). Methods The models of chronic epilepsy in rats were produced by sub - eclampsia dose of PTZ. The drug group received intragastric ganoderm lucidum spores powder infusion while injecting with PTZ intraperitoneally. The latency period and duration of epileptic seizures were recorded. The immunohistochemical staining was used to measure the immune reactivity of brain NF- κB/P65 protein. Results Under each high power microscope sight ( × 400), the level of NF-κB/P65 protein in hippocampus and brain cortex for epileptic model group was higher than that for normal control group ( P 〈0.01). And compared with epileptic model group, the latency period of epileptic seizures for drug group was longer on days 17, 21 and 25 ( P 〈0.05 or P 〈0.01), the level of NF-κB/P65 protein in hiplxxzampus and brain cortex for drug group was lower than epileptic model group ( P 〈 0.01 or P 〈0.05). Conclusion NF-κB may be involved in the occurrence and development of PTZ - induced epilepsy, ganoderma lucidum spores powder can down- regulate the expression of NF-κB, inhibit immune inflammatory reaction and decrease the excitability of nervous system, so it has protective effects on nerve cells.
