实验性变态反应性脑脊髓炎大鼠iNOS的动态表达及其意义

摘要 【目的】研究实验性变态反应性脑脊髓炎(EAE)大鼠诱导型一氧化氮合酶(iNOS)的表达规律及其在EAE发病中的作用。【方法】用豚鼠全脊髓匀浆(GPSCH)制备EAE模型。实验分为正常组、模型组(M组)。对实验动物进行神经功能评分检测,并在疾病初期、高峰期、缓解期三个时段,HE染色观察大鼠脊髓腰膨大区域炎性细胞浸润、免疫组织化学方法观察脊髓iNOS表达。【结果】模型组大鼠疾病初期腰膨大即有iNOS阳性表达,疾病高峰iNOS阳性表达达高峰,疾病缓解期iNOS阳性表达下降。【结论】EAE大鼠脊髓中iNOS阳性表达的细胞数量与疾病的活动性有关。推测iNOS的过度表达可能参与了EAE发病机制。 [Objective]To study the expression of inducible nitric oxide synthase(iNOS) in every pathological stage of experimental allergic encephalomyelitis(EAE) rat and to explore the its role in pathogenesis of EAE.[Methods]The EAE rat model was established with the homogenate of GP's spinal cord.The female Wistar rats were randomly divided into two groups: control group(n=10) and natural model group(M group,n=30).Model group was also divided into initial stage group,peak stage group,paracme group by the neurofunctional score.Infiltration of inflammatory cell was observed by HE staining method of lumbar intumescentia of the rat.The iNOS expression was measured with the immunohistochemistry method.[Results] The iNOS was expressed in the spinal cord of model in every pathological stage of EAE,and the number of iNOS cell of peak stage group was most among that of initial stage group and the paracme group(P〈0.05).[Conclusion]There is the expression of iNOS in the spinal cord of EAE rats,and the number of iNOS cell can be concerned with the disease reactiveness.It is presumed that overexpression of iNOS may participate in pathogenesis of EAE.
