β3GalT7基因转染HL-60细胞及对其生物学行为的影响

摘要 目的:初步探讨人类β1,3半乳糖基转移酶7(β3GalT7)基因对HL60细胞的细胞周期、侵袭能力等方面的影响。方法:通过脂质体介导将本实验室构建好的β3GalT7真核正义表达载体pEGFP-C1-T7-Sense和反义表达载体pEGFP-C1-T7-AntiSense转染入HL60细胞;流式细胞仪分析各细胞株的细胞周期改变;Transwell侵袭实验检测β3GalT7对HL60细胞侵袭转移能力的影响。结果:随着β3GalT7表达的增高,G2+S期细胞增多,且细胞侵袭能力增强;而转染反义表达载体pEGFP-C1-T7-AntiSense的结果是相反的。结论:过度表达β3GalT7的HL-60细胞株体外增殖和侵袭能力增强。 Objective: To explore the effects of β3 GalT7 gene on cell cycle, invasive potential of HL- 60. Methods: Transfect the recombinant plasmids pEGFP-Cl-T7-Sense and pEGFP-Cl-TT-AntiSense into HL-60 with liposomes. Cell cycle and invasive potential of the transfected cells with flow cytometry and transwell plate, respectively. Results: As the up-regulation expression of β3GalTT, the cell growth rate increased. At 48 h after transfection, significantly increased invasiveness was noted in HL60 transfected with pEGFP-Cl-T7-Sense. Conclusion: Over-expressing/β3Gal77 in HL-60 enhanced proliferative and invasive potential.
