差异凝胶电泳技术寻找冠心病心绞痛血瘀证的相关蛋白 被引量:9

Study on proteome of angina blood stasis syndrome by 2D-difference gel electrophoresis and mass spectrometry

摘要 目的:寻找冠心病不稳定性心绞痛血瘀证相关蛋白,探索其蛋白质组学特点。方法:采用DIGE差异凝胶电泳和串联质谱对8例冠心病不稳定性心绞痛血瘀证患者血瘀证程度变化前后及健康人的血浆进行蛋白质组学的比较研究,寻找冠心病不稳定性心绞痛血瘀证的相关蛋白。结果:初步发现Clusterin、ApoA-I在冠心病不稳定性心绞痛血瘀证患者的血浆中,表达低于健康人,血瘀证程度变轻后表达增加。而Fibrinogen β chain、Vitamin D-Binding Protein、Haptoglobin β chain、Haptoglobin α2 chain在冠心病不稳定性心绞痛血瘀证患者的血浆中,表达高于健康人,血瘀证程度变轻后表达降低。结论:Clusterin、ApoA-I、Fibrinogen β chain、Vitamin D-Binding Protein、Haptoglobin β chain、Haptoglobin α2 chain可能与冠心病不稳定性心绞痛血瘀证相关。 Objective: To seek differentially expressed plasma proteins of unstable angina blood-stasis syndrome. Methods: 2-dimensional diffirence gel electrophoresis and mass spectrometry were used to screen the differentially expressed plasma proteins among unstable angina blood-stasis syndrome and healthy volunteers. Results: Compared with healthy people, the express of Clusterin, ApoA-Ⅰ of unstable angina blood-stasis syndrome patient decreased while the expression of Fibrinogen β chain, Vitamin D-Binding Protein, Haptoglobin β chain, Haptoglobin α2 chain increased. Conclusion: Fibrinogen β chain, Fibrinogen γ chain, Haptoglobin β chain, Haptoglobin α 2 chain, Clusterin, ApoA-Ⅰ may be correlative with bloodstasis syndrome.
