摘要
目的制备聚乳酸一羟基乙酸共聚物(polylactide-co-glycolide,PLGA)包裹meta-四-(间羟基苯基)二氢卟吩[meta(tetrahydroxyphenyl)chlorin,m-THPC]的纳米型光敏剂,在体外研究其对人肝癌细胞光动力治疗(photodynamic therapy,PDT)作用。方法采用o/w的方法制备PLGA-m-THPC纳米微球,扫描电子显微镜下检测其粒径大小。荧光光谱仪检测其在含10%胎牛血清的细胞培养液中的吸收光谱。在人肝癌细胞株HepG2细胞的培养液内,分别加入不同浓度的PLGA-m-THPC,加红光照射后,观察细胞形态变化,并用细胞计数的方法测定细胞的存活率。结果PLGA-m-THPC纳米颗粒平均直径为180nm。PLGA-m-THPC在含血清培养液中的吸收光谱图显示其在652nm处有一个较强的吸收峰,并随着时间的推移吸光度增强。PLGA-m-THPC进入细胞主要聚集在胞浆,其对HepG2细胞的光敏杀伤作用随浓度的增加而增强。结论PLGA-m-THPC具有缓释、生物相容性好及光毒性低等优点,是一种理想的光敏剂。
Objective To produce polylactide-co-glycolide (PLGA) nanoparticles that encapsulate the photosensitizer meta (tetrahydroxyphenyl) chlorin (m-THPC) and to study the killing effect of photosensitizer PL- GA-m-THPC with red light irradiating on human hepatic cancer cell line Hep G2 in vitro. Methods The PL- GA-m-THPC were prepared by o/w multiple emulsion volatilizing method. Their general properties were observed. Absorbance spectra of PLGA-m-THPC in RPMI-1640(supplemented with 10% fetal bovine serum) were obtained on a spectrofluorimeter. A series concentration of PLGA-m-THPC solution were added to inoculum of human hepatic cancer cell line Hep G2. After irradiate with red light, the morphologic change and the survival rate of hepatic cancer cell were observed. Results The mean diameter of PLGA-m-THPC nanoparticles was 180 nm. It was found the absorbance spectra of PLGA-m-THPC at 652 nm had a peak and it increased with the changes of times. High fluorescence intensity of PLGA-m-THPC was localised mainly in the cytoplasm. And the photo- dynamic effect of PLGA-m-THPC on Hep G2 cell increased along with the concentration of PLGA-m-THPC. Conclusion PLGA-m-THPC can be used as a effective PDT nanoagent for the enhanced permeability, biocom- Dati-bilitv and low Dhototoxic.
