双酚A对鲫雌激素受体表达和雌二醇水平的影响 被引量:6

摘要 运用定量PCR技术和免疫荧光法研究了内分泌干扰物双酚A(BPA)对雌鲫的雌激素效应。以不同浓度的BPA[质量分数分别为1 mg/kg,50 mg/kg,100 mg/kg(注射剂量/鱼体重)]对雌鲫进行腹腔注射,然后分别于24 h和48 h检测肝脏中的α型雌激素受体(ERα)mRNA表达以及血清雌二醇(E2)水平。结果表明1 mg/kg和50 mg/kg BPA处理组ERαmRNA水平与对照组相比在48 h内均没有明显变化;100 mg/kg BPA处理组ERαmRNA水平在注射后24 h显著上调(P<0.05),48 h时仍维持较高水平。1 mg/kg BPA处理组E2水平在24 h显著下降(P<0.05),48 h回升至正常水平;50 mg/kg和100 mg/kg BPA处理组E2水平在24 h显著增加(P<0.01),48 h时仍维持较高水平。结果表明,BPA具有雌激素样效应,高浓度的BPA能够诱导雌鲫肝脏ERαmRNA表达和血清E2水平的上升。 The effects of bisphenol-A (BPA) on female Carassius auratus were investigated in vivo. Fishes were given intraperitoneal injection with the dose of I mg/kg, 50 mg/kg, 100 mg/kg of BPA. The expression of estrogen receptor alpha (ERa) mRNA in liver and the level of E2 in serum were checked in 24 h and 48 h after injection. The results showed that there was no significant increase of expression of ERoL mRNA both in 1 mg/kg and 50 mg/kg of BPA groups ; in 100 mg/kg of BPA group, the expression of ERoL mRNA increased significantly both in 24 h and 48 h (P 〈 0.05 ). The level of E2 decreased significantly in 1 mg/kg of BPA group in 24 h, then went up to the normal level in 48 h. In 50 mg/kg and 100 mg/kg of BPA groups, the levels of E2 increased significantly in 24 h and maintained at the high level in 48 h. These results indicated that BPA had estrogen-like effects. 100 mg/kg of BPA can up-regulate the expression of ERα mRNA in liver and the level of E2 in serum of female C. auratus.
