摘要
目的:探讨活性氧在微囊藻毒素-LR(MCLR)致细胞毒性中的作用。方法:不同剂量的MCLR作用HepG2细胞后,测定乳酸脱氢酶漏出率来反映细胞毒性,用荧光法和电子自旋共振技术分别检测细胞内总活性氧和羟自由基水平。结果:HepG2细胞经MCLR处理后,LDH漏出率呈剂量—反应式增加。抗氧化剂过氧化氢酶明显减轻MCLR诱导的细胞毒性。MCLR引起细胞内总活性氧及羟自由基水平升高。结论:MCLR增加了细胞内ROS的形成,羟自由基可能在MCLR引起的细胞毒性方面起着重要作用。
Objective: To study the role of reactive oxygen species (ROS) in cytotoxicity induced by microcystin-LR (MCLR). Methods: HepG2 cells were treated with MCLR, then the cytotoxicity was measured by the LDH release assay and the levels of ROS were determined by using fluorescent probes and electron spin resonance (ESR). Result: MCLR dose-dependently increased LDH release, which was inhibited by catalase. Significant increase in hydroxyl radical (OH) and ROS levels were detected in MCLRtreated cells. Conclusion: MCLR induced the generation of ROS, particularly hydroxyl radical may contribute to cytotoxicity of MCLR.
