摘要
以聚乙烯醇(PVA)为载体将从热带假丝酵母菌制备的粗酶进行固定化,并通过响应面方法对生物质乙醇的发酵工艺进行了一系列研究。在单因素试验的基础上,确定粗酶量、底物浓度和pH值为影响因子,应用Box-Behnken中心组合进行3因素3水平的试验设计,以乙醇得率为响应值,进行响应面分析。结果表明,PVA法固定化热带假丝酵母粗酶的最佳条件为:粗酶量0.36g,底物浓度8%,pH4.53;在该条件下进行固定化发酵的乙醇得率为25.84%。
The rude enzymes produced by Candida tropicalis were immobilized using polyvinyl alcohol (PVA) for entrapment. The production process of ethanol from corn straw biomass was studied with response surface analysis. Based on the single factor experiments, three independent variables, namely amounts of rude enzyme, substmte concentration and pH, were selected as impact factors. The experiments were arranged with 3 factors and 3 levels according to Box-Behnken central composite experiment design principles, and analyzed by response surface analysis using ethanol yield as response value. The results showed that the optimum condition for PVA immobilization of rude yeast enzymes were as follows: amounts of rude enzyme 0.36 g, substrate concentration 8% and pH 4.53. Under above conditions, the ethanol yield by immobilization fermentation was 25.84%.
