藏药佐太对秦艽中龙胆苦苷药物动力学的影响

Effect of Tibetan medicine Zuotai on the pharmacokinetics of gentiopicrosede in Gentiana

目的探讨藏药佐太对秦艽中龙胆苦苷在家兔体内的药动学特征的影响。方法对照组家兔给予龙胆苦苷提取液、实验组家兔给予龙胆苦苷和佐太的混合溶液后,采用RP-HPLC法测定给药后其血浆中的龙胆苦苷,DAS2.0软件计算各组家兔血浆中龙胆苦苷的药动学参数。结果对照组、实验组的家兔血浆中主要药物代谢动力学参数T1/2分别为1.66±0.32、1.74±0.39h,AUC0-t分别为99.84±28.36、86.35±4.53μg·h·ml-1,Cl分别为0.46±0.13、0.50±0.09L·h-1·kg-1,Vd分别为1.10±0.39、1.24±0.25L·kg-1,MRT分别为2.88±0.16、2.83±0.13h。两组的主要药动学参数无显著性差异。结论单次给予佐太对家兔龙胆苦苷的药动学特征无影响。

OBJECTIVE To study the effect of Tibetan medicine Zuotai on the pharmacokinetics of gentiopicroside in Gentiana. METHODS After control group of rabbits and experimental group of rabbits were administrated respectively with gentiopicroside solution and the mixture of gentiopicroside and Zuotai solution. The plasma concentration of gcntiopicroside was determined by RP - HPLC, and plasma concentration - time data were analyzed by DAS 2.0 software to get the related pharmacokinetic parameters of gentiopicroside. RESULTS The main pharmacokinetic parameters t1/2 of control and experimental groups were 1.66 ± 0. 32 h and 1.74 ±0. 39 h,respectively. AUC0-1 were 99.84 ±28.36 μg.h.ml^-1 and 86. 35 ±4. 53μg.h.ml^-1. Cl were 0. 46 ±0. 13 L.h^-1.kg^-1 and 0. 50 ± 0. 09 L.h^-1.kg^-1. Va were 1.10 ±0. 39 L.kg^-1 anti 1.24 ±0. 25 L.kg^-1. MRT were 2.88 ±0. 16 h and 2. 83 ±0. 13 h. The main pharmacokinetic parameters was not significantly different between control and experimental groups. CONCLUSION When rabbits were administrated with a single dose,the Zuotai could not significantly affect the pharmacokinetics of gentiopicroside in rabbits.
