摘要
目的建立测定罗布白麻叶中芦丁和槲皮素含量的方法。方法采用HPLC法,色谱柱为ZorbaxSB-C18(250mm×4.6mm,5μm),流动相为甲醇-0.04%磷酸(40:60),流速为1.0ml·min-1,检测波长为360nm,柱温为35℃。结果芦丁和槲皮素的线性范围分别为0.01~0.20μg和0.007~0.099μg;r分别为0.9999和0.9998;平均回收率分别为102.1%(RSD=2.0%,n=6)和99.5%(RSD=1.4%,n=6)。结论所建方法快速、简便、准确、重复性好,可作为罗布白麻叶药材质量控制的有效方法。
OBJECTIVE To develop a method for determining rutin and quercetin in A. hendersonii Hook f. METHODS HPLC method was adopted. Agilent Zorbax SB - C18 ( 250 mm×4. 6mm, 5 μm) column and methanol - 0. 04% phosphoric acid (40 : 60) as mobile phase were used. The flow rate was 1.0 ml.min^-1 and the detection wavelength was 360 nm. The column temperature was 35℃. RESULTS The peak areas vs the concentrations of rutin and quercetin showed good correlation in the ranges of 0. 01 - 0. 20 μg and 0. 007 - 0. 099 μg(r =0. 9999 and 0. 9998) , respectively. The average recoveries was 102. 1% for quercetin (RSD =2. 0% , n = 6) and 99. 5% for rutin (RSD = 1.4% , n = 6) ,respectively. CONCLUSION The method is simple, rapid and accurate.
