BALB/c裸鼠脾内移植人源性永生化肝细胞HepLL的生物学特性 被引量:1

摘要 目的研究人源性肝细胞HepLL经脾内途径移植后在体内的生物学特性。方法将人源性肝细胞HepLL移植入切除肝左叶和未切除肝左叶的BALB/c裸鼠脾内,通过病理和免疫组化方法检测其生物学特性。结果新建的人源性肝细胞移植入BALB/c裸鼠脾脏后可长期成团存活、增殖并表达肝细胞特异性标记物HEP,且有糖原的堆积,同时始终未检测到甲胎蛋白的表达。至移植后第30天,脾脏内移植的HepLL细胞切除左肝组比未切左肝组增生活跃。移植入脾脏的人源性肝细胞可向宿主肝脏移行并进入肝实质,同时可长期在宿主肝脏内增殖。结论移植入脾脏的人源性肝细胞HepLL可保持其原有的生物学特性,并能移行、整合入肝脏;肝左叶切除造成的肝细胞再生微环境有利于移植的人源性肝细胞HepLL的增殖。 Objective To investigate the biological characteristics of immortalized human hepatocyte (HepLL) after transplantation in the spleen of BALB/c nude mice. Methods The HepLL cells were transplanted into the spleen in BALB/c nude mouse with left hepatic lobe resected or reserved. The biological characteristics of HepLL cells in spleen were examined by pathology and immunohistochemistry methods. Results The HepLL cells survived in cluster for a long period after transplantation. The cells maintained ability of proliferation, expression of the specific marker HEP, and restoration of glycogen, while the expression of AFP was negative. Thirty days after transplantation the proliferation ability of HepLL cells was stronger in group with left liver lobe resected than that in group without resection. Pathological examination showed that the transplanted HepLL cells migrated and integrated into the host liver; and the integrated cells proliferated in the host liver for a long period. Conclusion HepLL cells transplanted into spleen maintain their biological characteristics and resection of the left liver lobe facilitates the proliferation of HepLL cells.
