摘要 目的探讨壳聚糖纳米颗粒对细胞免疫的调节作用。方法用50nm左右的壳聚糖颗粒与人外周血单个核细胞共同孵育,用3H-TdR掺入法检测纳米级颗粒对人单个核细胞的促增值作用;用CTLL-2细胞株检测白细胞介素-2(IL-2),用细胞病变抑制法检测干扰素α(IFN-α)。结果50nm的壳聚糖纳米颗粒在80-320μg/ml浓度下对人外周血单个核细胞有促增殖作用(P<0.05),并产生IL-2及IFN-α。结论壳聚糖纳米颗粒对人外周血单个核细胞有促增殖作用,促使其分泌细胞因子IL-2及IFN-α,引起Th1细胞免疫。 Objective Discussion chitosan nano particles to cellular immunity control action. Methods we used about 50nm chitosan particles and peripheral blood mononuclear cells to hatch together,detect the promotion of increment of nanoparticles to peripheral blood mononuclear cells by 3H-- TdR incorporation; detect IL-2 by CTLL-2 cell strain,detect IFN-α by cytopathic effect inhibition. Results 50nm chitosan particles can increase the quantity of peripheral blood mononuclear cells (P〈 0.05),and produce IL-2 and IFN-α. Conclusion Chitosan nano particles at concentrations of 80-320μ g/ml can promote proliferation of peripheral blood mononuclear cells; Chitosan nano particles can stimulate peripheral blood mononuclear cells producing IL-2 and IFN-α and activating Th1 cellular immunity.
