C-met蛋白和α平滑肌肌动蛋白在肝纤维化大鼠部分肝切除后肝组织中的表达

摘要 目的:研究C-met蛋白和α平滑肌肌动蛋白(α-SMA)在肝纤维化大鼠部分肝切除后肝组织中的表达情况及两者之间的关系。方法:成年雄性SD大鼠正常组、肝纤维化组和肝纤维化大鼠部分肝切除组(PH)。免疫组织化学、免疫荧光组织化学和免疫印迹法检测各组大鼠肝组织中C-met蛋白和α-SMA的表达情况。结果:肝纤维化大鼠部分肝切除后肝组织中C-met蛋白和α-SMA的表达都呈现出先降低后升高的规律,两者的变化具有高度的相关性。结论:C-met蛋白和α-SMA在纤维化肝再生过程中的表达都呈现出先降低后升高的规律性变化,肝星状细胞活化可能是纤维化肝再生过程中C-met蛋白表达的重要影响因素。 Objective: To research the expressions of C met and a smooth muscle actin (α-SMA) in hepatic tissues and their relationship after partial hepatectomy of rats with hepatic fibrosis. Methods: Adult male rats were divided into a normal group, a hepatic fibrosis group and the group of hepatic fibrosis with partial hepatectomy (PH). 12 hours, 1 day, 3 days, 5 days, 7 days and 14 clays after operation, the liver tissues were obtained from the PH group. Immtmohistochemistry, immuno- fluorescence histochemistry and Western blot were used to examine the expressions of α-SMA and C-met in liver tissue of every group. Results: After partial hepatectomy of rats with hepatic fibrosis, both the expressions of C-met and α-SMA in liver tissues decreased to the lowest point gradually, and then they increased gradually, there being a conspicuous relationship between their varia- tions. Conclusion: The expressions of both C-met and α-SMA decrease initially and then began to increase during the regenerative process of livers of fibrosis. Hepatic stellate cell may be a key factor affecting the expression of C-met during this process.
