奇正消痛贴膏对大鼠皮肤超微结构的影响和透皮吸收机理的研究 被引量:22

Effect of Qizheng Xiaotong Plaster on Epidermal Ultrastructure and the Mechanism of Transdermal Absorption in Rats

摘要 目的:阐明奇正消痛贴膏快速透皮吸收的作用机理。方法:选用Wistar大鼠,于大鼠背部皮肤涂搽药剂,应用透射电镜观察该药对大鼠皮肤超微结构的影响。结果:用药后使皮肤角质层细胞间距明显增大,细胞结构变得疏松,外层细胞脱落。该药进入角质层细胞内,与细胞内基质相作用,引起基底角质层肿胀和水化程度增加。停药后3~5d,表皮角质层可基本恢复正常。结论:奇正消痛贴膏可明显降低皮肤对药物的屏障作用,并使基底角质层水化程度增加,从而增加了透皮吸收。 Objective: To explore the mechanism of quick transdermal absorption of Qizheng Xiaotong Plaster (QXP). Methods: Skin of back in Wistar rats was smeared with medicine. Transmission electron microscopy (TEM) was applied to observe the ultrastructural changes of skin. Results: The space between cells in stratum corneum was widened significantly; the cellular structure became loosing, with enveloping cells desquamated. The drug was able to penetrate into the stratum corneum cells and interacted with intracellular matrix, resulting in stratum corneum swelling and higher hydration degrees. The structure of stratum corneum generally recovered 3--5days after drug withdrawal. Conclusion: The drug obviously lowers the barrier function of skin toward other medicines and increases hydration of basal stratum corneum, which improves transdermal absorption.
