亚低温对高血压病脑出血后水通道蛋白-4表达的影响

摘要 目的探讨亚低温对高血压病脑出血后水通道蛋白-4(AQP-4)表达的影响。方法建立高血压病脑出血大鼠模型,随机分为常温组和低温组,其中低温组又分出血后2、4、8、12小时四个时间点分别维持亚低温4小时和8小时,同时设正常对照组。取出血部位前后2mm以内的脑组织作标本,做AQP-4免疫组化染色、AQP-4和S-100双重染色及脑含水量测定,同时观察动物行为学变化。结果常温组脑含水量为(89.32±0.34)%,低温组出血后2小时含水量为(81.42±0.58)%,低温组出血后12小时与常温组含水量无显著性差异;常温组AQP-4的表达最多(吸光度0.27±0.04),低温组出血后2小时表达较少(吸光度0.17±0.02),两组间具有显著性差异(P<0.05),低温组出血后12小时与常温组间无显著性差别;常温组死亡率为50%,低温组出血后12小时为20%,其他组无死亡。结论早期亚低温可有效控制高血压病脑出血后脑水肿。 Objective To investigate the effect of hypothermia on Aquaporin-4(AQP-4) expressions after hypertensive cerebral hemorrhage. Methods Male Wester rats were feed with high salt and fat for 2 months and made cerebral hemorrhage with blood from rats tails. The rats were randomly divided into normothermia group and hypothermia group. Hypothermia group was divided into 8 groups, which included the rats received 4-hour or 8-hour period of hypothermia (32℃) 2,4,8,12 hours after blooded. AQP-4 was detected by immunohistochemistry and double staining. Brain water content and changes of animal behavior were observed as well. Results Normothermia group and 12 hours after blooded hypothermia group had severity edema. In the contrast, other groups conferred significant decrease edema (P〈0.05). Conclusion Early hypothermia treatment after hypertensive cerebral hemorrhage can effectively control cerebral edema.
