一氧化氮对大鼠主动脉血管平滑肌细胞骨桥蛋白表达的影响 被引量:1

摘要 目的观察一氧化氮(NO)对培养的大鼠主动脉血管平滑肌细胞骨桥蛋白表达的影响。方法用含10%小牛血清的DMEM培养液体外培养大鼠主动脉血管平滑肌细胞,随机分为对照组和不同浓度NO供体S-亚硝基-N-乙酰青霉胺(0.5、1、2、5mmol/L)干预组,应用反转录-聚合酶链反应及Westernblot技术结合光密度扫描分析,观察NO对血管平滑肌细胞的骨桥蛋白表达的影响。结果不同浓度NO供体S-亚硝基-N-乙酰青霉胺均明显抑制血管平滑肌细胞的骨桥蛋白mRNA和蛋白的表达,且具有剂量依赖性促进作用。结论NO能抑制血管平滑肌细胞骨桥蛋白的表达。 Objective To investigate the effect of Nitric oxide (NO) on osteopontin expression of rat aortic vascular smooth muscle cell(VSMC). Methods Rat aortic vascular smooth muscle cells were cultured in vitro with DMEM and 10% calf plasma. The cells were incubated with different concentration of S - nitroso - N - acetyl - d, 1 - penicillamine ( SNAP ) RT - PCR and western bloting analysis were used to observe the effect of NOon osteopontin expression of rat VSMC. Results Different concentration of SNAP could obviously reduce mRNA expression and protein synthesis of OPN, exhibited dose - dependently(P 〈 0. 05). Conclusion NO could inhibit osteopontin expression of rat VSMC.
