摘要
目的:探讨白细胞介素-4(IL-4)对巨噬细胞缺氧诱导促有丝分裂因子(HIMF)分泌及HIMF对血管平滑肌细胞增殖的影响。方法:用不同剂量的重组IL-4刺激培养的巨噬细胞,以逆转录聚合酶链反应(RT-PCR)检测巨噬细胞HIMF mRNA表达,荧光免疫组化激光共聚焦显微镜检测巨噬细胞HIMF蛋白表达;用终浓度分别为3×10-6mmol/L,9×10-6mmol/L,2.7×10-5mmol/L的重组HIMF刺激培养的平滑肌细胞,以3H-胸腺嘧啶核苷(3H-TdR)掺入法测定HIMF对平滑肌细胞增殖的影响。结果:Th2型细胞因子IL-4刺激巨噬细胞,HIMF mRNA及其蛋白明显表达(P<0.01);浓度为3×10-6mmol/L,9×10-6mmol/L,2.7×10-5mmol/L的重组HIMF明显促进平滑肌细胞增殖(与对照组比较P<0.01),且随着HIMF剂量的增加,促增殖作用增强。结论:Th2型细胞因子IL-4能刺激巨噬细胞表达HIMF mRNA及其蛋白,HIMF能促进体外培养的血管平滑肌细胞增殖。
Objective: To explore the effect of IL-4 on the expression of hypoxia-induced mitogenic factor (HIMF) of macrophages and HIMF on the proliferation of vascular smooth muscle cell (VSMC). Methods: Cultured macrophages collected from mouse peritonaeum were stimulated by different dose of recombinant IL- 4, the expression of HIMFmRNA and protein were detected by RT-PCR and laser confocal microscopy. The cultural VSMCs were stimulated with HIMF at different final concentrations of 3×10^-4mmol/L,9×10^-4mmol/L and 2.7×10^-5 mmol/L respectively. Then the effect of HIMF on the proliferation of VSMCs was detected by 3H--TdR. Results: When IL-4 stimulated macrophages, HIMFmRNA and protein expressed significantly (P〈0.01) ; HIMF promoted the proliferation of smooth muscle cell in a dose-dependent manner (HIMF groups compared with cantrol groups, P〈0.01).Conclusion.. IL-4 significantly induced expression of HIMFmRNA and protein in macrophages; HIMF promoted the proliferation of smooth muscle cells.
