摘要
目的:研究铜绿假单胞菌谷胱甘肽合成酶基因(gshb)突变体对MexXY-OprM外排泵表达的影响.方法:运用同源重组技术构建铜绿假单胞菌的谷胱甘肽合成酶基因(gshb)突变体,将连有MexXY-OprM外排泵启动子的pMS402质粒电转化到此突变体中,通过pMS402质粒上的Lux报道子发光值大小检测MexXY-OprM外排泵的表达变化情况.结果:MexXY-OprM外排泵的表达在谷胱甘肽合成酶突变体中显著降低.结论:铜绿假单胞菌中的谷胱甘肽对MexXY-OprM的表达具有促进作用,这将为进一步探讨MexXY-OprM的分子机制提供理论依据.
AIM:To study the impact of glutathione synthetase (gshb) gene mutant of Pseudomonas aeruginosa on the expression of MexXY-OprM efflux pump. METHODS : Using the homologous recombination technique we constructed the gshb mutant of P. aeruginosa and cloned the promoter regions of MexXY-OprM efflux pump into pMS402 and transferred it to the gshb mutant. The expression of MexXY-OprM was determined through the measurement of light production of the lux-based reporters on pMS402. RESULTS: The expression of MexXY-OprM was remarkably decreased in gshb mutant. CONCLUSION: Glutathione in P. aeruginosa can increase the expression of MexXY-OprM efflux pump; this result provides a theoretical basis for investiga- ting the mechanisms involved in MexXY-OprM.
