摘要
目的探讨臭氧氧化预处理通过诱导热休克蛋白70(HSP70)的合成,保护大鼠肾脏缺血再灌注损伤的作用与机制。方法建立原位大鼠单侧肾缺血再灌注动物模型,I/R前15d经直肠吹入氧气和臭氧的混合气体5.0~5.5ml(臭氧浓度50mg/L,1mg/kg体重,每日1次)。全自动生化分析仪检测尿素氮(BUN)、肌酐(Cr),比色法测定血清的脂质过氧化产物丙二醛(MDA)、超氧化物歧化酶(SOD)。Western blot检测HSP70蛋白的含量;逆转录聚合酶链反应(RT—PCR)方法检测HSP70的表达。结果。肾缺血再灌注24h后,血清中BUN、Cr、MDA明显增高,肾组织内HSP70表达明显增强(P〈0.05),经臭氧氧化预处理后,血清中的BUN、Cr、MDA均降低,SOD升高;HSP70表达升高更加明显(P〈0.05)。结论臭氧氧化预处理可以诱导大鼠肾缺血再灌注组织中HSP70表达,减轻大鼠肾脏缺血再灌注损伤。
Objective To investigate the protective effect of ozone oxidative preconditioning (OzoneOP) on heat shock protein 70 (HSP70) expression in renal ischemia-reperfusion (I/R) injury and the mechanisms. Methods A rat I/R model was established,and the renal OzoneOP was induced by rectal insufflation, 1 mg/kg, 15 ozone treatments, once every day, 5-5.5 ml at an ozone concentration of 50 mg/L. The parameters of renal function were determined by auto-biochemical analyzer. The contents of malondialdehyde (MDA) and superoxide dismutase (SOD) were measured by using chromometry. HSP70 was examined by RT-PCR and Western blot. Results Compared with the sham group, the expression of HSP70 in renal tissues and the serum levels of BUN, CR and MDA were increased significantly in the rats undergoing I/R procedure. After OzoneOP, the levels of serum BUN, Cr, MDA were significantly decreased, and SOD and HSP70 were significantly increased. Conclusion OzoneOP can increase the HSP70 expression and alleviate kidney I/R injury in rats.
