他莫昔芬对大鼠腹主动脉瘤形成的抑制作用 被引量:1

The effect of abdominal aortic aneurysm in the rats by treatment with tamoxifen

摘要 目的观察他莫昔芬对大鼠腹主动脉瘤(AAA)形成的影响及作用机制。方法40只雄性Wistar大鼠随机分成2组,实验组他莫昔芬预给药7d(每天15mg/kg体重),建立AAA灌注模型。术后7d切取腹主动脉。计算腹主动脉直径变化率,苏木素-伊红(HE)染色观察动脉壁炎性细胞浸润,Verhoff染色观察动脉壁弹性纤维,免疫组织化学观察动脉壁基质金属蛋白酶(MMP)-2,9及巨噬细胞的表达。结果他莫昔芬明显抑制AAA的形成,抑制率95%~99%。动脉壁内炎性细胞浸润明显减少,弹力纤维破坏受到抑制,MMP-2,9及巨噬细胞的表达受到抑制。结论他莫昔芬通过抑制大鼠腹主动脉壁内早期的炎症反应和MMPS的表达,抑制大鼠AAA的形成。 Objective To investigate the effect of tamoxifen on the formation of abdominal aortic aneurysm(AAA). Methods AAA model was established by perfusion of pancreas elasticity proteinase in 40 rats,which were divided into control team and Tam team. Tam teams were treated by tamoxifen, 15 mg/kg/d, to last a week. Results The treatment by tamoxifen inhibited AAA formation by 95% -99%. Infiltration of inflammatory cell and destruction of elastic fibers were evidently restrained. MMP-2,9 and macrophage activity were evidently restrained in the aortas of the tamoxifen-treated group. Conclusion Tamoxifen inhibit the development of AAAs in rats and reduce the expression of MMPs and the infiltration of inflammatory cell in aortic wall.
