摘要
目的观察反义转化生长因子(TGF)-β1质粒对肌腱粘连的治疗作用。方法将24只成年新西兰大白兔随机分成3组,行中趾屈指肌腱切断吻合术,腱鞘内分别注入生理盐水、反义TGF-1脱氧寡核苷酸和反义TGF-β1质粒,4周后取出肌腱进行生物力学测试;48只成年新西兰大白兔随机分成2组,行中趾屈指肌腱切断吻合术,腱鞘内分别注入生理盐水或反义TGF-β1质粒,于术后7、14、28、56d取出肌腱,原位杂交方法测定肌腱TGF-β1和Ⅰ型胶原mRNA的表达。结果生物力学结果表明使用反义TGF-β1质粒能有效的改善屈指肌腱术后的粘连;原位杂交显示反义TGF-β1质粒能有效的降低每个时间点TGF-β1和Ⅰ型胶原的表达。结论反义TGF-β1质粒能有效的抑制TGF-β1在肌腱损伤修复中的作用,减少粘连的形成。
Objective To investigate the preventive effect of antisense TGF-β1 recombinant plasmid on flexor tendon adhesion formation. Methods Twenty-four aduh New Zealand White rabbits were divided into three groups randomly after anastomonsis operation of flexor tendons:normal saline group,TGF- β1 oligonucleotide group, antisense TGF-β1 recombinant plasmid group. Tendons were harvested at 4th week and biomechanics testing was conducted. Fouty-eight adult New Zealand White rabbits were divided into two groups randomly after anastomonsis operation of flexor tendons: normal saline group, antisense TGF-β1 recombinant plasmid group. Tendons were harvested on the postoperative day 7,14,28 and 56 and the mRNA expression of TGF-β1 and Collagen Ⅰ was detected by in situ hybridization. Results The results of biomechanics testing indicated that antisense TGF-β1 recombinant plasmid could prevent adhesion formation of flexor tendon effectively after anastomonsis operation. The in situ hybridization examination revealed that TGF-β1 and Collagen Ⅰ mRNA expression in antisense TGF-β1 recombinant plasmid group was lower than that in normal saline group at each time point. Conclusion Antisense TGF-β1 recombinant plasmid can inhibit the function of the TGF-β1 effectively after the flexor tendon injured and repaired and prevent adhesion formatin.
