解毒通络注射液对MCAO大鼠脑组织炎性细胞因子的影响 被引量:5

摘要 目的:研究解毒通络注射液(JDTL)对局灶性脑缺血大鼠脑组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、细胞间黏附分子-1(ICAM-1)以及单核细胞趋化蛋白(MCP-1)含量的影响,探讨解毒通络注射液减轻脑缺血性损伤的作用机制。方法:成年雄性SD大鼠,线栓法复制大鼠局灶性脑缺血模型(MCAO),随机分成假手术对照组、模型组、解毒通络注射液高、中、低剂量组(83.2,41.6,20.8 mg.kg-1)和尼莫地平组。应用放射免疫法(RIA)检测脑组织匀浆TNF-α和IL-1β含量,酶联免疫吸附法(ELISA)检测脑组织匀浆ICAM-1和MCP-1含量。结果:脑缺血6,24 h,大鼠脑组织TNF-α,IL-1β,ICAM-1以及MCP-1含量均有不同程度的升高。解毒通络注射液高、中剂量组可以明显阻抑缺血脑组织上述细胞因子含量的增加。结论:解毒通络注射液对抑制脑缺血后炎症因子和黏附分子的表达具有良好的作用,能够减轻炎性损伤,阻抑脑缺血损伤级联反应,减轻缺血对神经元的损害。 Objective: To stydy the protective mechanism of Jiedu Tongluo (JDTL) injection on experimental cerebral ischemia. Method: Radioimmunoassay (RIA) was utilized to identify the content of TNF-α and IL1-β in brain tissue of rats suffered from 6 h and 24 h ischemia with MCAO, Enzyme-linked immunoadsordent assay (ELISA) was used to observe the level of MCP-1 and IC- MA-1. Result: After 6 h and 24 h of ischemia, the increased contents of TNF-α, ILI-β, MCP-1 as well as ICMA-1 was observed. JDTL injection dramatically inhibited the increase of TNF-α, IL1-β after 6 h and 24 h of ischemia. Conclusion: JDTL injection had inhibiting effect on TNF-α, IL1-β, MCP-1 as well as ICMA-1 caused by cerebral ischemia, the result suggested that JDIL injection can protect cerebral tissue from ischemic injury by inhibiting inflammatory factors.
