摘要
毛细管电泳法首次用于甜菊叶的絮凝工艺分析中,以斯替维苷(简称St)含量为检测指标,研究了实验参数对分离检测的影响。在含有23%乙腈和7%甲醇的50mmol/L硼砂缓冲液(pH=10.10)中,St和其他糖苷组分在15min内得到良好的分离。St在0.06—10.0mg/mL范围内呈良好的线性关系,检出限为0.02mg/mL。该方法已成功地运用于甜菊叶絮凝工艺中样品的检测。
A new capillary electrophoresis method was used in the study on flocculation technique of stevia extraction.The effects of modifiers, concentration and pH of running buffer,separation voltage and injection time were investigated.The best separation conditions are 50mmol/L borate buffer (pH=10.10),containing 23% acetonitrile and 7% methanol as modifiers.Under the optimum conditions,St could be separated from others within 15min.The calibration curves had good linearity in the range of 0.06—10.0mg/mL with the detection limits of 0.02mg/mL.The method was successfully used in the analysis of stevia samples of flocculation technique.
