摘要
本文主要探讨了利用生物芯片与高通量筛选技术研究中药作用机制的思路和方法。用人类蛋白芯片和细胞芯片,研究确定中药成分作用的靶标;并用靶标分子提取、分离相应的中药药效成分;再用中药药效成分作用于带有靶标分子的细胞或相关机体,通过基因芯片和抗体芯片检测细胞基因谱表达活性、细胞表面分子和分泌分子表达活性或机体血清活性分子含量等,并通过高通量筛选系统分析并得出结论,从中药有效成分作用的靶标定位及其对细胞、器官、系统乃至机体不同层面的功能状态影响,研究中药的作用机制。
Study how biochip technology was used to research the mechanism of chinese medicine. Human protein chip and cell chip were used to study target molecule of Chinese medicine effects. The target molecule was used to separate the active ingredient of Chinese medicine. The active ingredient of Chinese medicine acted on cell or organization which contained target molecule of Chinese medicine effects. Gene chip and antibody chip were used to detect expression of cell gene, secect molecule and cell surface molecule. And then high throughput screening was used to analyze data and conclusion was maken to study the effect of the active ingredient of Chinese medicine on cell, organization and system which contained target molecule of Chinese medicine effects.
