摘要
研究选用EE0.6基因和线粒体DNA控制区(D-loop)作为分子标记,用于合肥野生笼养东方白鹳的性别鉴定和遗传多样性检测。研究中成功地鉴定了东方白鹳成体和幼体的性别,并且双引物的引入克服了性别鉴定中假阴性。在合肥野生动物园28个笼养个体中共检测出11种单倍型,其单倍型多样性(h)为0.8598±0.0419,核苷酸多样性(π)为0.01035±0.0058,但单倍型在繁殖个体间分布严重失衡,子一代个体的单倍型主要集中在H1,H2,H3。因此,建议在进行圈养东方白鹳的繁殖管理时,注重提升各单倍型的奠基者作用,以保护笼养群体的遗传多样性。
EE0. 6 gene and mtDNA D-loop region were selected as molecular markers to execute determination of sex and genetic diversity research for oriental white stork in captive of Hefei wild zoo. We successfully identified the sex of mature and young oriental white storks based on EE0. 6 gene and eliminated the false negative bands by using two pair of primers. Based on the analysis of 463 bp of mtDNA D-Loop region, genetic diversity of 28 captive oriental white storks in Hefei Wild Zoo was investigated. A total of eleven haplotypes were identified in the captive colony. The haplotype diversity (h) was 0. 8598 ± 0. 0419 and nucleotide diversity (π) was 0. 01035 ± 0. 0058. However, the haplotype was not symmetrical distribution in samples. Most individuals of the first generation shared the haplotype of H1, H2 and H3. Therefore, we suggest that we should pay more attention to the haplotype founders so as to preserve the genetic resource of captive breeding colony.
