酸碱胁迫对蚬壳花椒组培苗内源激素的影响 被引量:6

摘要 为了研究不同酸碱度对蚬壳花椒组培苗的影响,在MS基本培养基上进行了pH值分别为2、3、4、6、8、10、11.5等不同酸碱度的胁迫处理,用酶联免疫法(ELISA)测定其在不同处理下的吲哚乙酸、脱落酸、赤霉素、玉米素核苷这4种内源激素含量的变化.结果表明:组培苗在pH值为3~8的范围内基本能正常生长,而pH值为2、10、11.5的处理明显受到伤害;4种内源激素含量对于不同的胁迫处理均呈现出一定的变化,pH值为6时,吲哚乙酸含量高于其它处理,其变化与脱落酸含量变化呈相反的趋势;玉米素核瞢的含量在2~3d时达到高峰,处理后期pH值为6的处理含量最高;赤霉素含量总体上呈波浪状上升的趋势. In order to find out the effects of acid and alkali stress on the contents of endogenous hormones in Zanthoxylum dissitum Hemsl, we treated its tissue culture seedlings on MS media with such different pH values as 2, 3, 4, 6, 8, 10 and 11.5, and assayed the content change of such four endogenous hormones as IAA, ABA, GA3 and ZR by using ELISA. The results show that the tissue culture seedlings can grow normally on the media within the range of pH values 3-8 but suffer obviously with the pH values 2, 10 and 11.5; that the four endogenous hormones all take on certain changes under different stress treatments, and the content of IAA in pH 6 is higher than that of other endogenous hormones, thus indicating that other treatments all have various degrees of inhibition to the growth of tissue culture seedlings; that the change rule of IAA content is just on the contrary to that of ABA, the content of ZR reaches its peak level after being treated for 2~3 d, and its highest content appears after the treatment with the medium of pH 6; and that the general content of GAs increases in a fluctuating way.
