摘要
目的:探讨高血压的神经免疫调节机制和滋阴潜阳方对高血压大鼠淋巴细胞内信号转导作用的可能环节。方法:采用酶联免疫法检测三磷酸肌醇(Inositol-1,4,5-trisphosphate,IP3)活性的改变。采用免疫组织化学染色法检测"二肾一夹"高血压大鼠淋巴细胞内蛋白激酶C(protein ki-nase C,PKC)蛋白表达。结果:模型组大鼠淋巴细胞内IP3活性和PKC蛋白表达明显增强,滋阴潜阳方对此有抑制作用。结论:滋阴潜阳方抑制淋巴细胞内IP3活性和PKC蛋白表达可能是其发挥神经免疫调节作用的机制之一。
Objective: To investigate neuroregulation and immunoregulation mechanisms for hypertension, and explore the potential pathway associated with the effect of Ziyin Qianyang Recipe (ZYQYR) on signal transduction of lymphocytes in rats with hypertension. Methods: The rat hypertension model was established by two-kidney-one-clip method. The content of Inositol-1,4,5-trisphosphate (IP~) in lymphocyles was measured by the method of enzyme linked immunosorbent assay. The expression level of protein kinase C (PKC) in lymphocytes was measured by immunohistochemical staining assay and DP801 Metamorph Image Analysis System. Resuhs:The level of IP3 and the expression level of PKC increased significantly in model groups compared with those of normal group and sham-operation group. ZYQYR had an inhibitive effect on the increment of PKC and IP3. Conclusion. It might be one of neuroregulation and im munoregulation mechanisms that ZYQYR inhibits the level of IP3 and the expression of PKC.
