摘要
目的观察中药加味补肝汤对实验性糖尿病大鼠坐骨神经脑源性神经生长因子(Brain-derived neurotrophic factor,BDNF)表达的的干预作用,探讨其对实验性糖尿病大鼠周围神经病变的防治作用。方法用1%链脲佐菌素诱发糖尿病模型,分别于治疗后第4周、8周末2个时间点处死大鼠,用MS302多媒体生物信号记录分析系统测定神经传导速度,用免疫组织化学法检测坐骨神经中BDNF蛋白的表达,通过Motic Images Advanced彩色图文分析系统测定免疫反应产物BDNF蛋白的灰度值。结果模型组、甲钴铵组、加味补肝汤组大鼠的神经传导均明显慢于正常对照组(P<0.05),甲钴铵组、加味补肝汤组快于模型对照组(P<0.05),加味补肝汤组与甲钴铵组比较无统计学意义。②模型组坐骨神经中BDNF阳性的表达比正常组明显减少(P<0.05),第8周时BDNF表达比4周时增多。甲钴胺组、加味补肝汤比模型组增多(P<0.05),但甲钴胺组与加味补肝汤组比较差异无统计学意义(P>0.05)。结论加味补肝汤能上调糖尿病大鼠坐骨神经中BDNF的表达,对糖尿病大鼠周围神经病变的有一定的防治作用。
Objective To observe the intervention effects of Jiaweibugan decoction on expression of BDNF protein in sciatic nerve of experimental diabetic rats, and to explore the preventive and therapeutic effects of Jiaweibugan decoction on peripheral neurophathy in experimental diabetic rats. Methods The diabetic rats were induced by 1% streptozotocin. The speed of conduction in nerve was recorded and the expression of BDNF protein of sciatic'nerve was detected by immunohisto- chemistry assay in the 4th and 8th weekends from the beginning of treatment respectively. The MS302 style of media biosignal system was used to record the speed of conduction in nerve, and computer image processing analytical technique was used to analyze immunohistochemical result by motic images advanced photochromic graphic processing system. Results (1) The velocity in model group was slower than that in normal control group( P 〈 0.05), and the velocity in cobalt ammonium group and Jiaweibugan decoction group was faster than that in model group(P〈 0.05), but there was no significant difference between cobalt ammonium group and Jiaweibugan decoction group. (2)The expression of BDNF protein in sciatic nerve of model rats was significantly lower than that in the normal control group(P〈 0.05), while that in the 8th week was higher than that in the 4th week. That in model rats treated by Jiaweibugan decoction and cobalt ammonium was significantly higher than that in those non-treated model rats(P〈 0.05), but there was no significant difference between cobalt ammonium group and Jiaweibugan decoction group. Conclusions Jiaweibugan decoction can increase the expression of BDNF in sciatic nerve of diabetic rats, and has preventive and therapeutic effects on peripheral neurophathy in experimental diabetic rats.
