摘要
利用靛蓝胭脂红褪色分光光度法测定葡萄糖氧化酶催化葡萄糖溶液产生的过氧化氢浓度,经过标准曲线转换得到葡萄糖氧化酶的活力。研究了缓冲液pH值、水浴温度、靛蓝胭脂红用量等对葡萄糖氧化酶活力测定的影响。结果表明:利用靛蓝胭脂红褪色分光光度法测定葡萄糖氧化酶活力时,缓冲液最佳pH值为4、最佳水浴温度为100℃、靛蓝胭脂红最佳用量为1.3mL。此方法的重现性较好,反应系统较稳定。
In this study, a novel method based on the fading of isatin is proposed to determine the glucose oxidase activity. The effects of buffer pH value, temperature of water bath and isatin dosage on determining process are analyzed. The results show that the best buffer pH value is 4, the best temperature of water bath 100 ℃ and the isatin dosage 1.3 mL. Besides, the method exhibits good repeatability and stability. Hence, it could therefore be a versatile method for determining the activity of glucose oxidase and would have great Promise in many fields.
