固相萃取快速分离高效液相色谱法测定决明子中的游离大黄素

Study on Determination of Emodin in Semen Cassiae by Solid Phase Extraction and Rapid High Performance Liquid Chromatography Method

目的:研究固相萃取快速分离高效液相色谱法测定决明子样品中的游离大黄素。方法:决明子样品提取液用Sep-Park-C18固相萃取小柱富集,以Agilent zobax C18(4.6 mm×50 mm×3.5μm)色谱柱为固定相,乙腈-0.1%醋酸溶液(50∶50)为流动相,流速为1 mL·min-1。结果:决明子中的大黄素在5 min左右达到基线分离;用紫外检测器检测,根据该色谱图的峰面积定量,标准回收率为101%-105%;RSD%为0.62%-1.53%。结论:用该方法测定了决明子中的游离大黄素,结果令人满意。

A rapid high performance liquid chromatography methodl HPLC ）for the determination of emodin in semen cassiae was tudied. Emodin can be enriched by solid phase extraction on a Sep-Park-C18 artridge, and separated on a Agilent zobax Eclipse XDB-C18 （4.6 mm×50 mm×3.5 μm）. The mobile phase consisted of acetonitrile-0.1%HAc（50：50）.The flow rate was 1.0mL min^-1. Emodin in semen cassiae was monitored by SPD, and determined by this method. The recovery of emodin was 101%-105%. The relative standard deviation was 1.28%-2.30%. The method was applied to the determination of emodin in semen cassiae with good result.
