兰州大尾羊脂肪组织总RNA提取方法的试验研究

Studies on Isolation Method of Total RNA from Adipose Tissue of Lanzhou Fat-tailed Sheep

在Trizol法的基础上通过优化操作规程、改进操作步骤等手段提取纯化了兰州大尾羊脂肪组织总RNA,并利用紫外分光光度计和凝胶电泳对其浓度和质量检测进行分析.结果表明:该方法可快速高效地获得适用于RT-PCR、基因表达的高通量分析、cDNA文库的构建等研究的高质量的总RNA.

Based on the Trizol, the experiment has extracted and purified the total RNA of Lanzhou fattail sheep by optimizing the operation process. And it also finished the detection and analysis of the concentration and quality of RNA by the ultraviolet spectrophotometry and Gel Electrophoresis. The results showed that the high quality total RNA can been got quickly and efficiently, which is used to RT- PCR, high throughput analysis of gene expression, the construction of cDNA library and so on.
