摘要
目的研究幽门螺杆菌Hp感染的胃癌和癌前病变中p53、ras基因蛋白的表达。方法美兰和W-S特殊染色方法确定Hp感染和免疫组化方法检测p53、ras基因蛋白表达。结果1.慢性萎缩性胃炎(CAG)、肠化生(IM)、异型增生(DYS)、胃癌(GC)中Hp感染率均高于浅表性胃炎(CSG)(P<0.05);2.在GC中p53,ras的阳性表达率显著高于IM(P<0.01),在IM中两者的阳性表达率要高于CAG(P<0.05);3.在IM中,Hp阳性组p53的阳性表达率高于Hp阴性组(P<0.05),在DYS和GC中,Hp阳性组p53、ras的阳性表达率均高于其阴性组(P<0.05)。结论Hp感染在胃粘膜病变的演变过程中起着重要作用,Hp感染与胃癌发生过程中的p53、ras密切相关,同时Hp可能作为促癌剂在胃癌的发生中起一定作用。
Objective To study the association between Helieobacter pylori (Hp) infection and the expression of p53, ras gene in gastric cancer and precancerous lesions. Methods Methylene blue staining and Warthin - Starry method were used to determine Hp. Immunohistochemical staining was used to detect the expression of p.53, ras gene protein. Results 1. The positive rate of Hp in chronic atrophic gastritis (CAG), intestinal metaplasia (IM), Dysplasia (DYS) and gastric cancer (GC) was significantly higher than that in CSG (P 〈0. 05 ). 2. The positive expression rate of p53, ras in GC was higher than that of IM in GC (P 〈 0. 01 ), the positive expression rate of p53, ras in IM was higher than that in CA(; (P 〈0.05). 3. The positive expression rate of p53 in IM with Hp positive was higher than that in IM with Hp negative ( P 〈0. 05 ). The positive expression rate of p.53 and ras in DYS and GC with Hp positive was higher than that in DYS and GC with Hp negative ( P 〈 0. 05). Conclusions Hp infection may play an important role in the development of gastric mucosal lesion. Hp infection was closely correlated with p53 and ras gene in the genesis of GC. Hp might act as a tumor promoter in the genesis of C,C.
