摘要
目的研究氯化甲基汞(MMC)对小细胞肺癌NCI-H446细胞周期的影响。方法采用流式细胞仪检测了MMC对NCI-H446细胞细胞周期的作用,并与目前最常用的化疗药顺铂(DDP)和同样具有剧毒的三氧化二砷(As2O3)做比较。结果MMC、DDP能使NCI-H446细胞表现为G0/G1期细胞数剂量依赖性明显增加,S期细胞数呈剂量依赖性明显减少;As2O3组细胞表现为明显的G2/M期剂量依赖性增加,S期细胞数剂量依赖性减少。结论MMC、As2O3、DDP均能诱导小细胞肺癌细胞株的细胞周期阻滞,但阻滞作用发生在细胞周期的不同时相。
Objective To investigate the influence of methylmercury (MMC) induced cell cycle stasis in human small cell lung cancer NCI-H446. Methods Flow cytometry FCM was used to examine the effects of MMC on the cell cycle of NCI-H446 cells and to compare with the current most commonly used chemotherapy drug cisplatin DDP and the samely highly toxic arsenic trioxide As203. Results MMC, DDP significantly increased G0/G1 phase cells with dose-dependent, significantly decreased S phase cells with dose-dependent; As2O3 group cells showed obvious G2/M phase increase and S reduction with dose-dependent. Conclusions MMC, As2O3, DDP all can induce cell cycle stasis in different phases of human small cell lung cancer NCI-H446.
