摘要
目的探讨葛根素对D-半乳糖模型小鼠记忆能力及胆碱能系统的影响。方法用Morris水迷宫法测试小鼠的空间记忆能力,用比色法测定大脑皮质胆碱乙酰转移酶(ChAT)、胆碱酯酶(AchE)活性,用碱性羟胺法测定乙酰胆碱(Ach)含量。结果与正常对照组相比,模型组学习记忆能力减退,大脑皮层ChAT、AchE活性降低,Ach含量下降。与模型组比较,葛根素治疗组学习记忆能力明显改善,大脑皮层ChAT活性提高,AchE活性无显著性差异,Ach含量提高。结论葛根素能改善D-半乳糖模型小鼠学习记忆能力和提高中枢胆碱能系统功能。
Objective To study the effects of puerarin on learning memory ability and cerebral cortex cholinergic system in D-galactose (gal)-lesioned mice. Methods Spatial memory ability was tested by Morris water maze, activity of choline acetyhransferase (CHAT) and acetylcholinesterase (AchE) in the cerebral cortex were determined by colorimetric method. And content of Ach was tested by alkaline hydroxylamine method. Results Model group had decreased learning and memory ability, reduced ChAT and AchE activities and Ach content compared with noimal control group. Puerarin group had improved learning and memory ability, increased ChAT activity in cerebral cortex and Ach content, but had no obviously changed AchE activity compared with model group. Conclusions The puerarin could significantly improve central cholinergic system function, and enhance the learning and memory ability in D-gal-lesioned mice.
