摘要
目的:研究芩连四物汤(川芎、当归、白芍、地黄、黄芩、黄连)中挥发油成分,并探讨其对催产素诱发小鼠离体子宫收缩效应的影响。方法:采用水蒸汽蒸馏法提取芩连四物方及其富含挥发油的当归、川芎组方药材的挥发油,并通过气相色谱-质谱联用技术对其分析鉴定和比较。结果:共鉴定成分70种,考察归一化计算面积大于0.5的29种成分,结果显示复方中的成分除(Z,Z-)9,12-Octadecadienoic acid外,均来源于药材,当归药材中除Cis-ocimene、γ-Cadinene、Aris-tolone成分在复方中未找到,其它均出现在复方中,川芎油中除Aristolone成分外,均出现在复方油中,藁本内酯是复方及组方药材挥发油中的主要成分。结论:对芩连四物方挥发油及其组方药材挥发油进行GC-MS成分比较分析;并提示藁本内酯可能是抑制离体子宫收缩的主要活性成分,为揭示芩连四物汤的效应物质基础提供依据。
AIM : To study the components of volatile oil of Qinlian Siwu Decoction (Rhizoma chuanxiong, Radix angelicae sinesis , Radix paeoniae alba , Radix rehmanniae , Radix scutellariae , Rhizoma coptidis , etc) and evaluate the activity of inhibiting the uterine contraction on mice in vitro. METHODS: The chemical compositions of essential oils obtained by steam distillation from Qinlian Siwu Decoction and its major constituting herbs ( Radix Angelicae sinensis, Rhizoma chuanxiong) , were analyzed by GC-MS. RESULTS: 29 components (70 in total) were recognized as the main ones from volatile oils of Qinlia Siwu Decoction. Its comparative peak areas came up to more than 0.5% of total. (Z,Z)-9.12-octadecadienoic acid were not found; and cir-ocimene, γ-cadinene and aristolone from Radix angeliae sinesis; aristolone came from Rhizoma chuanxiong. Ligustilide content was the highest one in volatile oils. CONCLUSION: The chemical components of volatile oil of Qinlian Siwu Decoction and its major constituting herbs (Radix Angelicae sinensis, Rhizoma chuanxiong) are comparatively analysed by GC-MS for the first time. The volatile oil of Qinlian Siwu Decoction shows antagonistic effect on the contraction of the isolated mouse uterus. Ligustilide are possible main active components.
