摘要
目的探讨大肠癌中唾液酸化的路易斯寡糖-X(Sialyl Lewis-X,SLeX)抗原和CD44v6基因蛋白的表达及临床意义。方法采用高敏感性的催化信号放大免疫组化技术,检测216例大肠癌、20例癌旁组织和20例正常大肠组织中SLeX抗原和CD44v6基因蛋白表达,分析与大肠癌临床病理因素的关系。结果SLeX抗原和CD44v6基因蛋白阳性表达率,大肠癌中分别为81.9%和83.3%,癌旁组织中分别为30%和35%,正常组织中均为阴性,三者之间差异具有显著性(P<0.01);SLeX抗原和CD44v6基因蛋白表达具有一致性,且与大肠癌的临床分期、分化程度、坏死、淋巴结及远处转移和预后有关(P<0.05)。结论SLeX抗原和CD44v6基因蛋白表达与大肠癌的侵袭、转移有关,并在大肠癌发生、发展中发挥重要作用,检测大肠癌中SLeX抗原和CD44v6基因蛋白表达可作为判定其预后的新的生物学指标。
Objective To explore the expression of Sialyl Lewis-X(SLeX) antigen and CD44v6 gene protein in colorectal cancer and its clinical significance.Methods High sensitive catalyzed signal amplification(CSA) immunohistochemical method was used to detect the expression of SLeX antigen and CD44v6 gene protein in 216 cases of colorectal cancer,20 cases of juxtacancerous tissues and 20 cases of normal tissues.The relationship was analyzed between the expression of SLeX antigen and CD44v6 gene protein and the clinicopathological individuals.Results The positive expression rate of SLeX antigen and CD44v6 gene protein in colorectal cancer were 81.9% and 83.3% and in juxtacancerous tissues was 30% and 35% respectively,with a significant difference between them(P〈0.01).There was a negative expression of them in normal tissues.The expression of SLeX antigen was consistent with that of CD44v6 gene protein in colorectal cancer and both of them were obviously correlated with clinical staging,degree of differentiation,necrosis,lymph-node metastasis and distant metastasis,and prognosis of the patients with colorectal cancer(P〈0.05).Conclusion There is a relationship between the positive expression of SLeX antigen and CD44v6 gene protein and invasion and metastasis of colorectal cancer,and the positive expression of them may play an important role in the development of colorectal cancer.Detecting the expression of SLeX antigen and CD44v6 gene protein may be considered as new biological indicators in judging the prognosis of patients with colorectal cancer.
