酸枣仁皂苷A对脑缺血再灌注损伤大鼠神经行为学及脑组织氨基酸含量的影响 被引量:5

摘要 目的:探讨酸枣仁皂苷A对脑缺血再灌注损伤大鼠神经行为学以及脑组织内氨基酸含量的影响。方法:先随机抽取6只大鼠作为假手术组,其余90只大鼠再随机分为缺血再灌注模型组、尼莫地平组和酸枣仁皂苷A组,各组再分设12h、24h、48h、4d和7d五个组,每个时间组6只大鼠。参考ZeaLonga线栓法制备大脑中动脉闭塞再灌注损伤模型,造模后分组给药并按时间点对大鼠进行神经行为学评分。之后,断头取右侧海马组织进行谷氨酸(Glu)及γ-氨基丁酸(GABA)含量测定。结果:尼莫地平及酸枣仁皂苷A组神经功能缺失症状较模型组有所减轻,Glu含量较模型组明显降低,GABA含量较之增加。结论:酸枣仁皂苷A在脑缺血急性期能抑制脑内Glu的释放,增加GABA含量,对大鼠神经功能缺失症状有一定改善,具有脑保护作用。 Objective:To study the effect of Jujuboside A on neuroethology during cerebral ischemia-reperfusion injury in rats and amino acid contents in their brain tissue. Methods:6 SD rats were randomly taken out as sham-operated group, and the rest 90 rats were randomly divided into cerebral ischemiareperfusion model group, Nimodipine group and Jujuboside A (JuA) group. Except thesham-operated group, every other group was divided into 5 timegroups as 12h, 24h, 48h, 4d, and 7d,with 6mice in each time group. The models of middle cerebral artery occlusion (MCAO) was established by Zea Longa's method. Each group was injected with related medicine and graded the neurological behavior on time-points. And then, removed the right side of hippocampus tissue for the determination of Glutamate (Glu) and Gamma-aminobutyric acid (GABA). Results:Compared with models group, the symptoms of rats neurological deficit were alleviating in Nimodipine group and JuA group, while the Glu contents in these two groups was obviously decreasing and GABA contents was increasing. Conclusion:JuA may inhibit the releasing of Glu, increase the contents of GABA in acute cerebral isehemia and improve the symptoms of rats neurological deficit to a certain extent, which has some protective effects on the brain.
