摘要 以永年大蒜为材料,采用生长量测定和根尖细胞有丝分裂观察等方法,初步研究了pH和NaCl胁迫对大蒜根、芽生长和细胞分裂的影响,旨在为选育耐盐碱的大蒜新品种奠定细胞学基础。结果表明,0.3%NaCl对大蒜根、芽的萌发和生长有一定的促进作用;0.7%和1.0%的NaCl胁迫对大蒜根、芽的生长表现明显的抑制作用,根部伸长区的细胞长度较对照的明显缩短,根尖细胞有丝分裂出现异常行为,如出现染色体桥、三极分裂和染色体解体等,1.5%NaCl完全抑制大蒜根、芽的生长;大蒜根、芽萌发和生长适合在pH6.5~7.5,低于pH6.5或高于pH8.5对大蒜根、芽萌发生长和根尖细胞分裂有一定影响。 The effect of different NaCl concentrations and pH values on garlic growth and cell division were studied by measuring the plant growth and observing the mitosis behavior.The results showed that 0.3% NaCl could stimulate the growth of garlic shoot and root in a certain degree,0.7% and 1.0% NaCl could restrain the garlic shoot and root growth obviously,and cause some abnormal cell division behavior during the mitosis,such as occurring chromosomal bridges,tri-pole spindle,chromosome disintegration and so on.,in addition,NaCl stress could significantly affect root cell elongation,suitable pH values for garlic growth were from 6.5 to7.5,and pH lower than 5.0 or higher than 8.5 could have some inhibiting to the plant growth and cell division.
