摘要 目的:探讨茶多酚对高糖条件下大鼠晶状体上皮细胞线粒体活性氧的影响。方法:体外培养大鼠晶状体上皮细胞,加入不同浓度的葡萄糖,加入不同浓度茶多酚进行干预,共聚焦显微镜检测各条件下大鼠晶状体上皮细胞线粒体活性氧量的变化,分析高糖及茶多酚干预对细胞线粒体活性氧表达的影响。结果:培养基中葡萄糖两种浓度(L1和L2组),大鼠晶状体上皮细胞(L)MTR、DCF荧光强度/μm2细胞面积L1组L2组比较,P<0.05。在30mmol/L葡萄糖浓度培养基中加入0.1μmol/L和0.3μmol/L浓度的茶多酚,大鼠晶状体上皮细胞死亡率CDR(%)及线粒体膜电位MMP(FI)L3组与L4组比较,P>0.05。L2组L3组及L2组与L4组比较P<0.05。结论:高糖条件下培养的大鼠晶状体上皮细胞线粒体活性氧产生增多,而应用茶多酚干预后细胞线粒体活性氧产生减少,细胞死亡率降低。 AIM :To study the effects of tea polyphenols (TP) on the expression of reactive oxygen species ( ROS ) in mitochondria of the rat 's lens epithelium under the condition of high glucose METHODS: The rat's lens epithelium cells cultured in vitro were intervened with different concentration of high glucose and TP. Changes of ROS in mitochondria of rat's lens epithelium cells in each condition were detected with confocal microscope to analyze the effects of high glucose and TP on the expression of ROS cells in mitochondria. RESULTS: The fluorescence intensity of MTR and DCF of the rat 's lens epithelium which cultured in vitro in different concentration high glucose had a significant difference between L1 and L 2 group (P〈0.05). Respectively added TP into the medium with 30mmol/L concentration of high glucose to get two groups of samples with different TP concentration, one is 0.1umol/L(group L3) and the other is 0.3umol / L(group L4). The results showed that the mortality of rat's lens epithelium cells and mitochondrial membrane potential were P 〉 0.05 between L3 and L4, P〈0.05 between L2 and L3, P〈0.05 between L2 and L4. CONCLUSION: ROS generated from the rat 's lens epithelium cells will increase under the condition of high glucose. Inversely, it will decrease after intervening with TP, and the mortality of lens epithelium cells will also decrease. These indicate that TP has the effect of inhibiting production of ROS in mitochondria and thus can delay apoptosis and decrease CDR of lens epithelium cells thereby.
