摘要
目的建立灵敏度高和特异性强的PCR方法检测广州管圆线虫几种中间宿主的感染。方法根据广州管圆线虫大亚基rRNA基因的序列特点,设计合成特异性引物,建立广州管圆线虫PCR检测方法,并对该方法的灵敏度和特异性进行验证。结果经琼脂糖凝胶电泳和DNA序列测定证实,建立的PCR检测方法具有极高的灵敏度和较好的特异性。结论初步建立的PCR检测方法可用于中间宿主——螺类携带广州管圆线虫的检测和野外疫情监测,具有良好的推广价值。
To establish a highly sensitive and specific a PCR assay for detection of Angiostrongylus cantonensis in several intermediate hosts, a pair of specific primers was designed and synthesized according to the sequence of large subunit rRNA gene, and the sensitivity and specificity of the method were investigated. The results of agarose gel electrophoresis and DNA sequence analysis indicated that the PCR method had high specificity and sensitivity. The developed PCR method can be used for detection of Angiostrongylus cantonensis in infected edible snail and for epidemic monitoring, thus it has great value in practical work.
