TRADD基因转染增生性瘢痕与瘢痕疙瘩成纤维细胞并诱导其凋亡的实验研究 被引量:4

摘要 目的探讨含有TRADD基因的重组腺病毒转染增生性瘢痕成纤维细胞与瘢痕疙瘩成纤维细胞,对纤维细胞凋亡特性的影响。方法利用分子克隆技术将TRADD基因转入Adeasy腺病毒载体中,经TNF预处理后,用重组腺病毒转染正常皮肤成纤维细胞、增生性瘢痕成纤维细胞与瘢痕疙瘩成纤维细胞,观察其对3种细胞凋亡率的影响。利用生长曲线、流式细胞仪检测转染前后3种成纤维细胞凋亡的情况。结果含有TRADD基因的腺病毒载体转染增生性瘢痕成纤维细胞与瘢痕疙瘩成纤维细胞后,细胞生长速度变慢,流式细胞仪显示细胞凋亡率增加。结论在TNF预处理的情况下,TRADD能提高增生性瘢痕成纤维细胞与瘢痕疙瘩成纤维细胞的凋亡率。 Objective To observe the effect of TRADD gene transfection on the apoptosis rate of hypertrophic scar fibroblasts and keloid fibroblasts. Methods An adenovirus vector which contains TRADD gene was constructed by the technology of molecular cloning, Then we transfected the adenoviruses into normal fibroblasts, hypertrophic scar fibroblasts and keloid fibroblasts which were treated by TNF. The method of MTT and the flow cytometer were used to test the proliferation rate and the apoptosis rate before and after the transfection. Resuits After the transfection, the growth rate of hypertrophic scar fibroblasts and keloid fibroblasts become slower. Conclusion The TRADD protein can increase the apoptosis rate of hypertrophic scar fibroblasts and keloid fibroblasts which were treated by TNF in vitro.
