The hemagglutination-inhibition (HI) assay is a traditional method for assessing immune responses to influenza virus. It is well known that the receptors for avian and human influenza virus on the host cells are different; therefore, HI test based on red ceils from various species of animals containing different receptors may have different sensitivity. In this study, The traditional chicken red cells-based HI ( cHI ) with horse red ceils based-HI (hHI ) methods was compared in the detection of antibodies against H5N1 influenza virus. It found that two kinds of method have different performances when 3 kinds of HI antigens were used, but the differences between two assays were roughly within the 2-fold range. Therefore, the traditional chicken red ceils-based HI is still suitable for the serological evaluation of pandemic influenza vaccines.
